4. 제목: Design, Fabrication and Application of Microfluidic Cell Culture Devices

In this talk I want to present a novel method of creating multi-compartment cell culture devices for biological research applications. Culturing and real time monitoring of cells in microfluidic device is a new challenge in the field of cell biology. Miniaturization of cell culture devices using elastomeric microfluidic devices offers unique advantages for the studies of development, migration, axon guidance, synaptogenesis, axon regeneration and programmed death of cells. Microfluidic cell culture devices were fabricated in PDMS, a transparent and biocompatible material, using soft lithography and replica-molding techniques reproducibly, precisely, and rapidly. The microfluidic devices were used for culturing human umbilical vein endothelial cells (HUVEC), MDA-MB-231 human breast cancer cells, NIH 3T3 mouse fibroblasts, and primary rat cortical and hippocampal neurons. Viability inside the devices is comparable and the morphology of the cells is equivalent to those in controls. Advantages of this chamber over standard culturing methods include increased control over neuronal microenvironments and less reagent usage. Microfluidics is used to maintain fluidic isolation within the chamber, allowing selective delivery of positive or negative stimuli to cells. This technique enables culturing of cells inside microfluidic devices for a number of biological research applications where cells need to be exposed to well-controlled fluidic microenvironment.
